چکیده

A glyceollin I accumulation o f about 6000 pmol • mg dry weight, a tenfold increase above control root tissue, was found in one type o f nodule from Glycine max which had been infected with a fix~ strain (61-A-24) o f Rhizobium japonicum. In nodules infected with one other ineffective (ßx~) strain o f Rhizobium japonicum (RH 31-Marburg) or with two f i x + strains o f Rhizobium japonicum (61 -A -101 and USDA 110) no increase in glyceollin I concentrations above control values was found at either 20 d or 34 d after infection. Nodules infected with Rhizobium japonicum 61-A-24 are distinguished by an early loss o f the peribacteroid membrane in the infected host cell, whilst the bacteroids themselves remain stable.

Soybean nodulin-26 gene encoding a channel protein is expressed only in the infected cells of nodules and is regulated differently in roots of homologous and heterologous plants.

Nodulin-26 (N-26) is a major peribacteroid membrane protein in soybean root nodules. The gene encoding this protein is a member of an ancient gene family conserved from bacteria to humans. N-26 is specifically expressed in root nodules, while its homolog, soybean putative channel protein, is expressed in vegetative parts of the plant, with its highest level in the root elongation zone. Analysis...

Nodulation, nutrient accumulation and yield of rainfed soybean in response to indigenous soybean-nodulating Bradyrhizobia in the Himalayan region of Kashmir-Pakistan

The use of efficient and effective nodulating Bradyrhizobia strains considered as anecologically and environmentally sound management strategy for soybean production. A 2-yr(2009 and 2010) field experiment was conducted to evaluate the effects of seven indigenousBradyrhizobium strains, one exotic TAL-102 and three N fertilizer rates, i.e., 25, 50 and 100kg N ha-1 on the productivity and N2 fixa...

Topology and phosphorylation of soybean nodulin-26, an intrinsic protein of the peribacteroid membrane

Soybean nodulin-26, a homologue of bovine eye lens major intrinsic protein (MIP-26), is an integral protein of the peribacteroid membrane in symbiotic root nodules. It comprises 271 amino acids with six potential transmembrane domains and lacks an amino-terminal signal sequence. A full-length nodulin-26 cDNA and its various deletion derivatives were transcribed in vitro after linking them to ba...

Iron Uptake by Symbiosomes from Soybean Root Nodules.

To identify possible iron sources for bacteroids in planta, soybean (Glycine max L. Merr.) symbiosomes (consisting of the bacteroid-containing peribacteroid space enclosed by the peribacteroid membrane [PBM]) and bacteroids were assayed for the ability to transport iron supplied as various ferric [Fe(III)]-chelates. Iron presented as a number of Fe(III)-chelates was transported at much higher r...

Leghemoglobin-derived radicals. Evidence for multiple protein-derived radicals and the initiation of peribacteroid membrane damage.

Reaction of H2O2 with ferric leghemoglobin (metLb, the monomeric, oxygen-carrying, heme protein from root nodules of nitrogen-fixing plants) has been previously shown to generate an iron(IV)-oxo (ferryl) species and at least one protein radical. The latter has been suggested to be a tyrosine-derived phenoxyl radical present at Tyr-133 in the soybean protein and Tyr-138 in the lupin protein. To ...
